Why use PACE™ technology for COVID-19 work?

Earlier in the Covid-19 pandemic, detection assays using the TaqMan technology were established and applied to mass testing, and this remains the standard approach.  However, sequencing of patient Covid samples has revealed new strains of the virus in circulation.

The detection of new Covid variants cannot be achieved without the development of new assays, which requires the design and validation of new probes and primers and the subsequent synthesis of large quantities of both.  The fluorescently-labelled probes required for TaqMan assays, aside from being expensive, are also in short supply because of huge demand.  The consequence of this demand is that oligo synthesis of new probes is very much delayed.

At 3CR Bioscience we think a more flexible screening approach is in order.  In contrast to TaqMan, the fluorescent reporting system of PACE-RT is universal and included in the master mix so there is no requirement for the purchase of labelled probes.  In addition, PACE-RT™ uses assays that are composed only of unlabelled primers so the design, validation and implementation of new assays is quick, easy and cost-effective.

Once assays are validated, their application to large-scale testing requires only the synthesis of primers, rather than probes.  We design assays free-of-charge for use with all PACE technologies, providing the primer sequences to the customer, whereupon the primers can be ordered from any preferred oligo manufacturer.  We provide PACE-RT master mix for use with PACE assays and we also provide PACE-RT based kits for HV69-70del and N501Y strains comprising PACE-RT master mix and assays.

Why use ProbeSure™ technology for COVID-19 work?

Our ProbeSure™ Covid-19 One Step RT-PCR Kit contains both the assays and master mix required for the real-time or endpoint detection PCR detection of RNA from the SARS-CoV-2 virus (identifying markers N1 & N2 with an RNase P control). As labs have different plate reading capabilities, we have produced three different iterations of the assays to allow users to choose the most convenient and time-efficient kit for their equipment.

Prices are for the complete kit which includes:

  • ProbeSure™ RT-PCR Master Mix (containing enzymes)
  • Primer sets for 3 markers – N1, N2 and RNase P (as recommended by the CDC)

Features of the kit include: 

  • Sensitivity as low as 1 copy per μL in a 5 μL reaction
  • Detection using real-time PCR to report generation in under 2 hours
  • Detection using endpoint PCR, forming distinct clusters enabling users to increase throughput
  • Multiplex kit, but singleplex and duplex options available
  • Assays used are for markers recommended by the CDC (US Centres for Disease Control) to target nucleocapsid regions of the SARS-CoV-2 genome
  • These assays target regions of the SARS-CoV-2 genome that are not present in other coronaviruses (e.g., SARS, MERS, bat SARS-like coronavirus)
